VP test
indicator (5% a_naphthol (3drops)+ 40 %KOH(1drop) to 0.5 ml culture

- ve :yellow color
عندي ملاحظة على تصوير سيت ال E. Coli و الكلبسيلا انتبهوا ال indole -ve و ال MR- , + VP في الكلبسيلا عكس ال E. Coli
ارجوا الانتباه الها كما موضح في الجدول

MacConkey agar:
Indicator: neutral red
Eosin Methylene Blue agar (EMB):
(contains methylene blue
Oxidase test
(1% solution of
oxidase dye: N, N, N, N-tetramethyl-p-phenylenediamine dihydrochloride)
Nitrate reduction test
.sulfanilic acid (5 drops).
- α-naphthol (5 drops).
**if no colorchangeAdd zinc powder (trace)
Triple Sugar Iron agar (TSI)
Indicator is phenol red
Indole test:
Indicator: Kovak's
Methyl red (MR)
Indicator: methyl red (3 drops of MR reagent + 2.5 ml culture).
Voges Proskauer (VP test)
Indicator: 5% α-naphthol (3 drops) + 40% KOH (1 drop) to 0.5
ml culture.
Urease test
Indicator: phenol red
