درس كبير بس مهم

لو هقيس B_ Quantitative measurements في الحالة دي هدخل عديد من ال Standard ومعاه العينات علي الجهاز وأعرف الجهاز العينات والاستاندر واعطي للجهاز قراءات ال Standard الجهاز يرسم Calibration curve between Absorbance and concentration of standard وبعد كده نسقط عليها باقي ال absorption بتاع العينات ويحصل علي باقي Concentration of samples خلي بالك في حالات ELISA العلاقة بين ال absorption and concentration علاقة طردية لكن ليست خطية يعني مثلا لو ال Concentration 100➡ absorption 0.1 Concentration 200➡ absorption 0.4 Concentration 300➡ absorption 0.5 Concentration 400➡ absorption 0.9 كده ال الزيادة في absorption غير ثابتة بزيادة ال concentration يعني Concentration /absorption isn't constant فنضطر إننا نعمل calibration curve.. نجيب كذا حالة معلومة التركيز standard.. هنا لازم كذا حاله بخلاف ال Spectrophotometer ممكن حالة واحدة standard لأن هناك علاقة طردية وخطية معا فالزيادة في absorption ثابتة بزيادة ال concentration يعني Concentration / absorption is constant ______ Types of ELISA : There are four types or kinds of ELISA tests: كل نوع له صورة بالأسفل، أنظر إليها 1_ Direct ELISA: attachment of an antigen to a polystyrene plate followed by an enzyme-labeled antibody that can react with the antigen and then substrate that can be measured الموضوع كله ⬅نضع العينة (الانتيجن اللي في الدم) في ال well،هيمسك في جدار ال well ب polystyrene ، هضيف عليه الأنتي بودي اللي في reagent هذا الأنتي بودي ماسك فيه الإنزيم، يقوم الأنتي بودي يمسك في الأنتيجن ، نحضن ونغسل، ثم نضع ال Substrate اللي هيشتغل عليه الإنزيم، يقوم الإنزيم يشتغل علي substrate ويتلون لو الأنتيجن موجود. EX : not widely used but commen in immuno-histochemical staining هي سريعة فعلا لكن مشكلتها ان ال الأنتي بودي المحمل بإنزيم من الممكن أن لا يتحد مع الأنتيجن وبالتالي تعطي نتيجة سلبية كاذبة فهو مش specific وكمان مش sensitive. 2_ Indirect ELISA: "indirect ELISA" refers to an ELISA in which the antigen is bound by the primary antibody which then is detected by a labeled secondary antibody.  الموضوع كله نضع العينة (الانتيجن اللي في الدم) في ال well،هيمسك في جدار ال well ب polystyrene، هضيف عليه الأنتي بودي اللي في reagent، يقوم يمسك فيه، نحضن ونغسل، ثم أضيف أنتي بودي آخر ماسك فيه الإنزيم، يقوم يمسك في ال Ab الأولاني ، نحضن ونغسل، ثم نضع ال Substrate اللي هيشتغل عليه الإنزيم، يقوم الإنزيم يشتغل علي substrate ويتلون لو الأنتيجن موجود. ملحوظة ⬅ممكن يكون المراد قياسه ال Ag اللي وضعناه في الأول أو Ab اللي وضعناه ثاني شيئ EX ➡ detection of infectious agents as HIV,HBV,HCV مميزاتها إن الأنتي بودي بيبقي موجه ل Mor epitopes in detected Ab ➡mor Ag Ab combination ➡ more substrate react with enzymes ➡ it gives mor intense colour ➡ test become more sensitive 3_ Sandwich ELISA:  الأشهر An antibody to a target protein is immobilized on the surface of microplate wells, then sample containing Ag is added, then detecting ( secondary) Ab added Direct ➡ detecting Ab is enzyme link Indirect ➡ detecting Ab isn't enzyme link, but secondary enzyme linked Ab added Substrate added ➡color changes هنا هنحط الأنتي بودي علي جدار ال well، ونضيف الأنتيجن اللي في الدم،نحضن ونغسل، وبعد كده نحشر الأنتيجن بأنتي بودي آخر زي الساندويتش If direct these antibody is labeled by enzyme, if indirect نحضن ونغسل، ثم نضيف انتي بودي آخر ماسك فيه الانزيم يقوم يمسك في الأنتي بودي الأولاني ،نحضن ونغسل ، ثم نضع ال Substrate اللي هيشتغل عليه الإنزيم فيتلون. EX : tumour markers, hormones,... فايدة عمل هذه الطريقة الساندويتش، إنها تزيد من Specificity of immunological interaction لأني بمسك الأنتيجن باتنين Ab ففي الغالب صعب يكون الإمساك خاطئ. sandawitch ELISA is more specific and sensitive. ملحوظة⬅ في الأنواع السابقة، كلما زاد الشيئ المراد قياسه كلما زاد اللون، بعكس النوع القادم كلما كلما زاد الشيئ المراد قياسه كلما قل اللون. NB ➡ In the previous types, the greater the object to be measured the more color, unlike the next type (competitive) , the greater the object to be measured the less color. 4_ Competitive ELISA:

1_ primary antibody incubated with sample 2_ antigen-antibody complexes form 3_ More antigen = more binding = less free antibody أول حاجة نجيب Ab ونحضنه مع العينة هيمسكوا معا ، وكل ما كان Ag كثير كل يمسك في ال Ab كثير ويقل ال Free Ab 4_ Mixture (inhibitory Ag) is added to the plates 5_ unbound Ab binds these inhibitory Ag 6_ wash away Ag-Ab complexes 7_ secondary Ab labelled with enzyme directed to inhibitory Ag and then substrate added 8_ More color change less Ag in sample بعد ذلك نضع الخليط ( inhibitory Ag) علي plate، هيمسك في ال free Ab ، نعمل wash ،بعد ذلك نضع secondary Ab labelled with enzyme directed against inhibitor Ag and then add Substrate More color change ➡ less Ag in sample طبعا ممكن تبحث عن أنتي بودي بنفس الطريقة. EX: T3,T4 and progesterone. ______ شوية ملحوظات حابب أقولها: 1_ وقت التحضين مهم جدا علشان الأنتيجن يتحد مع الأنتي بودي 2_ وقت ال wash مهم جدا علشان Removal of unbound antigen antibody 💿So Incubation and washing is much important for good results 3_ ELISA kits have 96 well microtiter plate 4_ measurement of Absorbance in ELISA occurs at 450 nm 5_ أهم انزيمين يستخدموا في ال ELISA A _ alkaline phosphatase act on p- NPP substrate to give chromagen product B_ Horse radish peroxidase act on TMP and H2O2 to give chromagen product 5_ may use stopping solution as NAOH or H2SO4 Stop Solution in ELISA is a solution used in the final steps to stop the action of an enzyme on a substrate and so stop any further color changes To standardize the procedure,All factors should be constant including volumes, concentrations and timing of reaction.. (ELISA) Enzyme Linke ImmunoSorbant Assay : بسم الله الرحمن الرحيم هتكلم اليوم إن شاء الله عن ال ELISA بداية حابب أشكر كل إنسان علمني أو أضاف ليا معلومة يوما ما، فعلا مش عارف أعملهم إيه لكن يكفيني الآن إني أقول لهم جزاكم الله خيرا، ومن قال ذلك فقد أبلغ في الثناء كما قال رسول الله صلى الله عليه وسلم روي الترمذي بإسناد صحيح عن أسامة بن زيد رضي الله عنهما قال: قال رسول الله صلى الله عليه وسلم: "من صنع إليه معروف، فقال لفاعله: جزاك الله خيراً، فقد أبلغ في الثناء".  ليس لي أيضا الا الدعاء لهم بظهر الغيب، عن ابن عمر رضي الله عنهما، قال: قال رسول الله صلى الله عليه وسلم: (ومن صنع إليكم معروفا فكافئوه، فإن لم تجدوا ما تكافئونه فادعوا له حتى تروا أنكم قد كافأتموه)). Basic principles : فكرة عمله بكل بساطة إني باعمل Recognition of antigen by antibodies ال Ab ماسك فيه enzyme Ab labelled with enzymes هذه الإنزيمات تعمل علي تحويل ال Colorless Substrate ➡ coloured product (chromagen) تحوله من عديم اللون إلي لون او حتي يحوله من لون لآخر هذا اللون ممكن بالنظر كده اعرف إذا كان إيجابي أم سلبي وممكن عن طريق جهاز ال Spectrophotometer اقيس Absorbance لكل حالة واعرف التركيز بالضبط. طبعا كل ما زاد تركيز الشيئ المراد قياسه هيمسك فيه الأنتي بودي أكثر وبالتالي يزداد الانزيم اللي ماسك فيه وبالتالي يعمل أكثر علي ال Substrate ويحوله للمادة الأخري ويظهر اللون أكثر، أشوفه ملون أكثر، أقيس تركيزه عن طريق جهاز ال Spectrophotometer هلاقي تركيزه أعلي لازم بين كل خطوة وخطوة أعمل Incubation and wash بعد كده أقرأ اللون بال Spectrophotometer Antigens from the sample are attached to a surface. Then, a further specific antibody is applied over the surface so it can bind to the antigen. This antibody is linked to an enzyme, and, in the final step, a substance containing the enzyme's  substrate is added. The subsequent reaction produces a detectable signal, most commonly a color change in the substrate. ______ Advantages of ELISA : صراحة لها مميزات كثيرة أهمها إنها حساسة ورخيصة ولا تحتاج لأدوات كثيرة ويسهل إجرائها سريعا ولا يوجد بها إشعاع 1_ very sensitive 2_ doesn't require specialized equipment 3_ avoid hazard of radioactivity 4_ to some extent cheap 5_ easy to perform and quick procedures. ______ Application of ELISA : ال ELISA لها تطبيقات كثيرة جدا في معاملنا من أهمها 1_ hormones eg HCG,TSH, .... 2_ Infections agents as bacteria, viruses, parasites fungal eg HCV, HCV,HIV,... 3_ tumour markers 4_ serum proteins 5_ Immunoglobulins as IgG,IgM,IgA 6_ detecting potential food allergens, such as milk, peanuts,  walnuts,  almonds, and eggs 7_ ELISA can also be used in toxicology as for certain classes of drugs. 8_ others....., أهم استخدامات هي الهرمونات والفيروسات ودلالات الأورام...... إلخ ______ Steps of ELISA : طبعا على حسب البامفلت، هنقول نوع معين وهو Direct sandawitch ELISA 1_ add samples into a well of the microplate strips (well coated by antibody against what's measured) 2_ add another antibodies with enzyme linked. ( usually HRP conjugate) 3_ incubate about for ... minutes then wash (repeated wash) 4_ add enzyme substrate to all well (CHROMAGEN, TMB,... ) 5_ incubate about for.... minutes. 6_ add stop solution 9_ بعد كده قيس اللون الناتج Read the Optical Density (O.D.) at.. nm using a microtiter plate reader within.. minutes. Calculation of results Standard curve is used to determine the amount in an unknown sample. لو هقيس A_ Qualitative measurement يبقي كده هعمل Positive and negative control and samples واحسب ال Cutoff من البانفلت طبعا ممكن بالنظر كده اعرف ال Positive and negative ولكن احنا بنستخدم ال Cutoff Cutoff value determined by adding a factor to the mean Absorbance of the negative or positive control value. وبحسب منطقة اسمها ال grey zone = + or - 10% of Cutoff value وبعدين نشوف الناتج Above grey zone is➡ positive result below grey zone is ➡negative result Reading with grey zone ➡retested in a duplicate ➡ if still reactive confirm by another methods
